High-throughput expression screening of histidinetagged proteins
The increased number of protein variants and mutants that need to be handled in drug development requires high-throughput methods for protein expression and purification. In this study, automated methods for expression screening, with parallel enrichment of affinity-tagged target protein are described*. A combination of His MultiTrap™ HP 96-well filter plates packed with Ni Sepharose™ High Performance medium and Tecan Freedom EVO ™ liquid handling workstation was used, followed by SDS-PAGE for final analysis.
