How should I prepare the cell suspension for single-cell dispensing?

Cell suspension protocol for single-cell dispensing
  1. Start by washing the cell in the flask with PBS two times, then add trypsin-EDTA. Rock back and forth to ensure trypsin-EDTA covers completely the cell monolayer.
  2. Proceed by incubating the cell flask with trypsin-EDTA for 10 minutes at 37 °C
  3. Remove flask from incubator and add 3 ml of complete media to neutralize trypsinization reaction
  4. Using a serological pipette, pipette up and down 10 times to mix the cell suspension
  5. Transfer 1 ml of cells to a sterile microcentrifuge tube
  6. Determine cell viability and concentration using a cell counting method (automated cell counter, hemocytometry, etc.)
  7. Normalize the concentration of your cells to 200 cell/µl
  8. Dispense the desired number of cells in each well with Uno
