Highly sensitive detection and characterization SARS-CoV-2 in nasal samples with Trio RNA-Seq™ library preparation kits

Our Trio RNA-Seq library prep kit detects more viruses and potential pathogens than other methods. More sensitive detection enables researchers to better determine the underlying causes of disease.

This method increases detection sensitivity from nasal swab samples and is the method of choice for IGATech. (Data has been generated and analyzed by IGATech in the framework of a research initiative coordinated by Istituto di Genomica Applicata and funded by ARGO, Italy.)

McClain et al. used Universal Plus mRNA-Seq to study the host’s transcriptional response of the peripheral blood system from COVID-19 patients. In this study, they were able to show a robust transcriptional response to SARS-CoV-2, which will enable researchers to identify key biological mechanisms of the disease.

Accurate, reproducible amplification from limited input samples with Ovation® RNA-Seq System V2 for high-quality cDNA generation

qPCR is a widely used tool for quick, high-throughput target validation for determining the presence or absence of pathogens in a sample. Low input amounts and low abundance of the target transcript reduce the effectiveness of standard RT-PCR methods. 

Based on the proven SPIA technology, our Ovation RNA-Seq System V2 provides increased qPCR detection sensitivity, enabling analysis of very low-abundance RNA viruses that are below detection levels of standard methods. Ovation RNA-Seq System V2 can be effectively integrated with the qPCR workflow to accurately and reproducibly interrogate hundreds of degraded samples, while more of the original precious sample can be archived and stored for later applications such as additional qPCR tests or NGS assays.

Recent peer-reviewed studies (published in 2020) demonstrate the successful application of our NGS library preparation and qPCR kits to study SARS-CoV-2 in patient samples:

  • Maurano, MT., et. al. from New York University successfully performed large scale genomic surveillance of 236 patient samples from the NYC metropolitan area. The researchers showed that there was undetected community spread and concluded that the majority of SARS-CoV-2 strains in NYC originated from Europe. Trio RNA-Seq was chosen over other kits for this study due to a lower rate of duplicate reads, especially for lower viral input samples.
  • Shen, Z., et. al. performed metatranscriptome sequencing for the bronchoalveolar lavage fluid of eight SARS-CoV-2 patients, 25 community-acquired pneumonia (CAP) patients and 20 healthy controls to study viral evolution and how the virus interacts with other microorganisms in the lung . The authors concluded that the virus evolves after infection, which may impact its virulence, infectivity, and transmissibility.
