What are the concentration limits?

Maximum stock concentrations that can be dispensed using the D300e

1. The concentration limits are:

  • 10 mM for small molecules up to 1,000 Da
  • 1/3 mg/ml (T8+ cassettes) or 0.3 mg/ml (D4+ cassettes ) for proteins up to 300 kDa, or DNA up to 10,000 base pairs

For T8+ cassettes, biomolecule concentrations up to 3 mg/ml can be dispensed. When using T8+ cassettes for bulk dispensing, where no dispensed volume is below 1 nl, the loading volume can be raised to 20 µl for biomolecule concentrations up to 1 mg/ml.

Please note that, if you are dispensing your sample in a mixture with other biomolecules (eg. albumin), the total concentration of biomolecules must be considered (eg. [sample] + [albumin]). The concentration of your small molecules, proteins or nucleic acids influences the viscosity of the liquid. At concentrations above the indicated thresholds, the viscosity impacts the dispensing performance.

2. Further information

You can find further information about what can be dispensed with the D300e in the ‘Dispensing software user guide’, which can be accessed from the D300eControl software through File > Documentation > Dispensing Software User Guide > Appendix E.
